Abstract:

COATSOME^® SS-Lipid, a novel lipid platform for nucleic acid delivery is composed of highly biodegradable, ionizable lipids. Rational engineering of functional groups of the lipid to improve biodegradation in the cytoplasm was shown to vastly improve gene expression while at the same time reducing hepatic toxicity. Here we detail the chemistry and biology of LNP-mRNA complexes and preclinical data for a CRISPR-Cas9 genome editing therapeutic and an mRNA vaccine program.
